Furthermore, human pepsin remains stable at pH 7.4 and can be reactivated by hydrogen ions from any source. Thus, most tap and bottled water (typically pH 6.7-7.4) does not affect the stability of pepsin. The purpose of these in vitro studies was to investigate whether artesian well water containing natural bicarbonate (pH 8.8) could irreversibly denature (inactivate) human pepsin and to determine its potential acid buffering capacity.

Methods: Laboratory studies were performed to determine whether human pepsin was inactivated by alkaline water at pH 8.8. In addition, the buffering capacity of alkaline water was measured and compared to the two most popular commercially available bottled waters.

Results: Alkaline water at pH 8.8 irreversibly inactivated human pepsin (in vitro) and its hydrochloric acid buffering capacity far exceeded that of normal pH water.

Conclusions: Unlike normal drinking water, alkaline water of pH 8.8 denatures pepsin immediately, rendering it permanently inactive. In addition, it has excellent acid buffering capacity. Thus, consumption of alkaline water may be therapeutically beneficial for patients with reflux disease.
